  3. Molecular Mechanisms of Pain in Chronic Pancreatitis”.  National Institute of Diabetes & Digestive & Kidney Diseases R01. $1,819,550.00/4yrs. Principal Investigator: Pankaj Jay Pasricha; Co-investigator (50% effort): Guang-Yin Xu; Funding: funded.  This proposal examines the hypothesis that the pathogenesis of pain in chronic pancreatitis involves changes in pancreas-specific sensory neuronal excitability and neurotransmitter expression/release and that such changes are mediated, at least in part, by nerve growth factor (NGF) expressed in pancreatic tissue. The goals of this project are to determine the ionic and molecular basis for increased excitability of pancreas-specific primary nociceptive neurons in chronic pancreatitis, to determine the effects of chronic pancreatitis on expression and release of peptide neurotransmitters and their role in maintaining nociceptive sensitization and to determine the role of NGF in the pathogenesis of pain behavior and pancreas-specific sensory neuronal responses in chronic pancreatitis.  Guang-Yin will perform pancreas-specific sensory neuron excitability and ion channel activity experiments.
