A different Protocol

Procedure
Grow cells overnight in YEPD.
Remove 200 l, add to 5 ml sterile water.
Spin 5 min.
Resuspend pellet in 2.5 ml SPM.
Incubate at 23C with good aeration for 2-4+ days and check under microscope for the formation of tetrads.


Recipes
zymolyase solution
0.5 mg/ml in 1M sorbitol:
5 mg zymolyase
10 ml 1M sorbitol

SPM
3 g potassium acetate
0.2 g raffinose
Mix in 1 L of sterile water
